I. Basis of Application

Laboratory headspace vials comply with GB/T 32465-2015 “Chemical Reagents—Headspace Sampling Vials for Gas Chromatography” and are applicable to the full process of standardized operations for 20 mL standard clamp-mouth/threaded headspace vials used in headspace analysis of VOCs in water, food, pharmaceuticals, and environmental samples.

II. Selection and Warehouse Acceptance Standards

Vial Material
For conventional volatile components: transparent borosilicate headspace vials; for light-sensitive or easily degradable components: brown light-shielding vials. Vial walls must be uniform, and the mouth must be flat with no chipping, cracks, or scratches. Damaged vials must be scrapped immediately and must not be used in experiments.
Sealing Component Compatibility
Use PTFE/silicone rubber composite seals exclusively, with the PTFE layer facing the interior of the vial; use high-temperature-resistant seals for high-temperature analysis. Clamp-mouth vials must be fitted with dedicated aluminum crimp caps, and threaded vials with corresponding threaded caps. Mixing different specifications of caps and seals is strictly prohibited.
Batch Control
Use the same model of headspace vials and seals for all experiments within the same batch; perform a blank validation on new batches of consumables before use; they may be used in batches only if no interference peaks are detected.

III. Cleaning, Drying, and Activation Standards

1. Pre-treatment of New Headspace Vials
1) Sonicate in pure water for 10 minutes to remove dust;
2) Dry in an oven at 120–150°C for 2 hours, then cool to room temperature; for trace analysis, purge with nitrogen at 150°C to remove residual release agents;
3) Do not load directly into the instrument to avoid blank baseline interference.
2. Cleaning Procedure for Reusable Vials
1) Empty residual liquid; degrease by ultrasonic cleaning in chromatographic-grade acetone or methanol for 15 min;
2) Rinse repeatedly with ultrapure water 3–5 times;
3) Dry at 105°C until completely cooled;
4) If crystalline residues are present on the vial mouth, wipe with a solvent before washing;
5) The use of dishwashing detergent or potassium dichromate solutions is strictly prohibited, as they may introduce impurities and cause ghost peaks.
3. Criteria for Scrapping
Vials with worn or deformed rims, stubborn stains on the inner walls that cannot be cleaned, cracks in the vial body, or septum puncture holes ≥3 times shall be scrapped immediately.

IV. Standardized Sample Loading Procedures

Sample Volume Control
Fill liquid samples to 1/3 to 1/2 of the vial’s volume, leaving sufficient space for vapor-liquid equilibrium; filling to capacity or overfilling is strictly prohibited to prevent failure of phase equilibrium, liquid leakage during heating, and significant data deviations.
Sampling Requirements
Insert the pipette below the liquid surface and add the sample slowly to minimize air bubbles; for volatile or easily oxidizable samples, perform the operation in an ice bath at low temperature, and seal the vial immediately after filling to minimize exposure to air.
Matrix Addition Guidelines
When adding salts, buffers, or internal standards, pour them slowly along the inner wall of the vial to avoid splashing onto the septum at the vial opening, which can cause contamination and adsorption losses.

V. Core Guidelines for Sealing and Capping (Key to Preventing Leaks)

Place the septum centrally in the cap, ensuring it is not offset, wrinkled, or damaged;
For clip-top vials, use a dedicated capping tool to apply even pressure; after capping, gently shake the vial to ensure the cap is secure and not skewed;
For screw caps, tighten until you feel a click, then turn an additional 1/4 turn; do not use excessive force that could deform the septum;
After sealing, invert the vial and let it stand for 30 seconds; check for leaks and ensure no bubbles escape;
The septum is a single-use consumable and must not be reused; replace it immediately after multiple punctures.

VI. Temperature Equilibration and Instrument Sampling Procedures

Place sample vials upright and securely in the headspace tray; do not tilt or invert them to prevent liquid from contacting the septum and leaching impurities;
Strictly adhere to the testing standards for equilibration temperature and duration; maintain consistent conditions for samples within the same batch;
The autosampler needle must penetrate vertically; avoid repeated punctures at the same location on the same septum;
Do not remove or shake the sample vials during the heating process, as this disrupts the gas-liquid equilibrium and leads to inaccurate quantification.

VII. Sample Storage and Management Guidelines

Label immediately after sealing: sample name, number, sampling time, and batch;
Store volatile samples refrigerated at 4°C away from light, in an area distant from organic solvents and reagent bottles;
Complete testing of water samples and VOC samples within 7–14 days; run unacidified samples within 24 hours;
Samples exceeding the storage period shall be discarded and may not be used for quantitative analysis in the People's Republic of China...

VIII. Daily Quality Control and Safety Requirements

Include full-procedure blank vials in each batch to investigate background interference from the vial itself, cleaning solvents, and the separator;
Check for proper sealing before high-temperature heating to prevent pressure buildup and splashing inside the vial;
Promptly empty and clean headspace vials after the experiment; store them by category, with clean vials and vials awaiting cleaning managed in separate zones;
Clean the capping machine regularly; be cautious of sharp edges on aluminum caps to prevent cuts during operation.

IX. Proper Handling of Common Issues

Significant differences in peak areas among parallel samples: Verify that the sample volume, cap tightness, and equilibration time are consistent across all samples;
Contaminant peaks in blanks: Re-bake the headspace vials to reactivate them and replace the seals with brand-new, low-background ones;
Leakage after temperature stabilization: Discard any headspace vials with damaged necks; replace all aged seals.
